Contrave is another experimental
diet drug being investigated by its maker Orexigen Therapeutics Inc. Their
stock symbol is OREX and traded on the NASDAQ. Contrave is an interesting medication
because like QnexaŽ, Contrave is actually just a combination of two drugs used for separate purposes. It is the
adding of Naltrexone IR and Bupropion SR in a unique combination. Although different
amounts of each drug making Contrave where studied, it was during phase
2 testing that the combination of 32mg of Naltrexone IR (Immediate Release) and
400mg of Bupropion SR (Sustained release) showed on average the better benefit
to risk ratio. The two drugs that make up Contrave are an interesting combination, Naltrexone is mainly prescribed for alcohol and drug addiction/abuse and often goes under the trademarked name of VivitrolŽ or ReviaŽ. Bupropion is an atypical antidepressant which is also used as a smoking cessation. Bupropion has also been prescribed off label to help weight loss but not approved as such. But when combined together with Naltrexone it is showing promise as a weight loss drug. Contrave has also shown favorable testing results in decreasing metabolic syndrome, help with diabetic patients to lose weight and can help with the obese and depression. Contrave was submitted as an NDA (New Drug Approval) to the FDA in hopes of having it approved March 31st, 2010. There also will be an advisory committee that will review Contrave before this date (tentative date December 7th, 2010) and give its opinion and recommendations to the FDA before they ultimately review and decide its fate January 31st, 2011. Contrave does exceed the FDA requirements for a drug to be considered a drug for obesity with average weight loss in phase 3 studies between 9 and 15%. However, it will be the side effects that most likely determine if it becomes shelved or not. Some of the more common side effects where considered mild to moderate in tolerance and consisted of things like Nausea, headaches, constipation and tended to happen more at the beginning of the study. I wonder if a medication such as Contrave if approved would hold more promise for the obese who also struggle from substance abuse. Looking at what this drug is created from, both ingredients have properties of helping out addictions of different kinds.
